چکیده

Butyrylcholinesterase (E.C. 3.1.1.8) is present in all vertebrates (1, 2). We present here the sequence of rabbit BCHE gene deduced from three genomic clones described elsewhere (3). Each clone contained one exon and together covered the entire coding sequence. The structure of the rabbit BCHE gene is identical to the human gene (4) and to Torpedo ACHE gene (5). Positions of splicing sites within the coding sequence are conserved but non coding sequences are not conserved except for a portion of the 3' end untranslated sequence which may have a role in the regulation of the mRNA. The identity with human BCHE is 90.3% in nucleotidse over the coding sequence and 91.6% in amino acids. The high proportion of bases A and T is a characteristic of both human and rabbit BCHE gene. Monoclonal antibodies to human butyrylcholinesterase reactive with butyrylcholinesterase in animal plasma.

Five mouse anti-human butyrylcholinesterase (BChE) monoclonal antibodies bind tightly to native human BChE with nanomolar dissociation constants. Pairing analysis in the Octet system identified the monoclonal antibodies that bind to overlapping and independent epitopes on human BChE. The nucleotide and amino acid sequences of 4 monoclonal antibodies are deposited in GenBank. Our goal was to det...

The Distribution of Cholinesterase

Acetylcholinesterase and butyrylcholinesterase activities were demonstrated in transverse sections of the rabbit ear artery using a “direct coloring” thiocholine technique. BW 284C5l, iso-OMPA and DFP were used as inhibitors, and acetyl and butyrylthiocholine iodides as substrates. Both enzymes caused stain deposits at the medial-adventitial border of the artery, in a region where sympathetic n...

Protective efficacy of catalytic bioscavenger, paraoxonase 1 against sarin and soman exposure in guinea pigs.

Human paraoxonase 1 (PON1) has been portrayed as a catalytic bioscavenger which can hydrolyze large amounts of chemical warfare nerve agents (CWNAs) and organophosphate (OP) pesticides compared to the stoichiometric bioscavengers such as butyrylcholinesterase. We evaluated the protective efficacy of purified human and rabbit serum PON1 against nerve agents sarin and soman in guinea pigs. Cataly...

Phylogenetic tree construction of butyrylcholinesterase sequences in life forms.

Butyrylcholinesterase is an enzyme with few known physiological functions. It is related to acetylcholine that was shown to be expressed in a variety of life forms. We performed a search using the human butyrylcholinesterase gene (HGNC:983;MIM:177400), and found the sequence in a broad spectrum including plants, bacteria and animals. Therefore butyrylcholinesterase appears to have evolved early...

Genotyping the butyrylcholinesterase in patients with prolonged neuromuscular block after succinylcholine.

BACKGROUND Succinylcholine remains the standard neuromuscular blocking drug for tracheal intubation in emergency situations. The short duration of action is due to its rapid hydrolytic degradation by butyrylcholinesterase (plasmacholinesterase). Multiple variants of this enzyme are known (A, F, S, H, J, K variants) with different effects on enzyme activity. This study was undertaken to evaluate...
